The encoded protein also stabilizes constitutive heterochromatin to maintain the overall chromatin structure. The active, hypophosphorylated form of the protein binds transcription factor E2F1. Experimental studies have shown that this variant disrupts mRNA splicing (PMID: 10882758). This variant has been observed in individual(s) with retinoblastoma (PMID: 10882758, Invitae). This variant is not present in population databases (ExAC no frequency). This sequence change affects codon 444 of the RB1 mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the RB1 protein. This variant also falls at the last nucleotide of exon 13 of the RB1 coding sequence, which is part of the consensus splice site for this exon. -
