The BRCA2 c.8158G>A (p.Glu2720Lys) variant has been reported in the published literature in at least one individual with prostate cancer (PMID: 21952622 (2011)). This variant has also been described to be located in a region of the BRCA2 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -

The p.E2720K variant (also known as c.8158G>A), located in coding exon 17 of the BRCA2 gene, results from a G to A substitution at nucleotide position 8158. The glutamic acid at codon 2720 is replaced by lysine, an amino acid with similar properties. This alteration has been reported in a cohort of individuals with prostate cancer who underwent testing for alterations in BRCA2; authors listed p.E2720K as an unclassified variant in this study (Kote-Jarai Z et al. Br. J. Cancer, 2011 Oct;105:1230-4). This amino acid position is well conserved through mammals but not in all available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
