BRCA2 (HGNC:1101): (BRCA2 DNA repair associated) Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The largest exon in both genes is exon 11, which harbors the most important and frequent mutations in breast cancer patients. The BRCA2 gene was found on chromosome 13q12.3 in human. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, May 2020]

Variant summary: The BRCA2 c.10127C>G (p.Ser3376X) variant results in a premature termination codon, predicted to cause a truncated or absent BRCA2 protein due to nonsense mediated decay, which are commonly known mechanisms for disease. However, this variant is located in the last exon in the gene, and other truncating variants located in this exon upstream of this variant have been classified as VUS or benign by our lab (p.Glu3316fsX2, VUS; p.Lys3326X, benign; p.Glu3342X, VUS; p.Ser3366fsX4, benign). One in silico tool predicts a damaging outcome for this variant. This variant is absent in 121162 control chromosomes. The variant of interest has not, to our knowledge, been reported in affected individuals via publications and/or reputable databases/clinical diagnostic laboratories; nor evaluated for functional impact by in vivo/vitro studies. Taken together, the functional significance of this truncating variant located in the last exon is unknown, therefore it has been classified as a VUS until additional evidence becomes available. -

However, this variant is located in the terminal exon of the BRCA2 gene and is not expected to result in non-sense mediated decay (NMD). In the published literature, this variant has been reported in individuals with non-small cell lung cancer (PMID: 34632253 (2021)) and prostate cancer (PMID: 36922933 (2022)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, we are unable to determine the clinical significance of this variant. -
