The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and isoleucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change alters the normal expression level and the distribution of protein in cells (PMID: 28877251). However, the clinical significant of this finding is unknown. This variant has been reported to segregate with bilateral pulverulent nuclear cataracts in multiple affected individuals from a single family (PMID: 28877251). This variant is not present in population databases (ExAC no frequency). - |
