The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. This variant has not been reported in the literature in individuals with FANCM-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with leucine at codon 191 of the FANCM protein (p.Pro191Leu). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and leucine. -
