MAP kinases, also known as extracellular signal-regulated kinases (ERKs), act as an integration point for multiple biochemical signals. This protein kinase lies upstream of MAP kinases and stimulates the enzymatic activity of MAP kinases upon wide variety of extra- and intracellular signals. As an essential component of MAP kinase signal transduction pathway, this kinase is involved in many cellular processes such as proliferation, differentiation, transcription regulation and development. This variant has not been reported in the literature in individuals with MAP2K1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with leucine at codon 306 of the MAP2K1 protein (p.Pro306Leu). The proline residue is moderately conserved and there is a moderate physicochemical difference between proline and leucine. -
