This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with pancreatic cancer (PMID: 33169439). ClinVar contains an entry for this variant (Variation ID: 484197). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies have shown that this missense change affects PALB2 function (PMID: 33169439). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.P104L variant (also known as c.311C>T), located in coding exon 4 of the PALB2 gene, results from a C to T substitution at nucleotide position 311. The proline at codon 104 is replaced by leucine, an amino acid with similar properties. This alteration was identified in an individual with a personal and family history of pancreatic cancer (Zhang Y et al. Hum Mutat, 2021 02;42:150-163). Functional studies showed performed on this variant showed reduced recruitment of PALB2 and RAD51 to nuclear foci and reduced HR activity (Zhang Y et al. Hum Mutat, 2021 02;42:150-163). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
