Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277). For these reasons, this variant has been classified as Pathogenic. This sequence change removes the last 3 nucleotides of exon 8, and affects a donor splice site in intron 8 of the TP53 gene. It is expected to disrupt RNA splicing and/or create a premature translational stop signal, and likely results in an absent or disrupted protein product. -
