Likely pathogenic, criteria provided, single submitter | clinical testing | Ambry Genetics | Jan 09, 2017 | The p.C275W variant (also known as c.825T>G), located in coding exon 7 of the TP53 gene, results from a T to G substitution at nucleotide position 825. The cysteine at codon 275 is replaced by tryptophan, an amino acid with highly dissimilar properties. This alteration is in the functionally critical DNA binding domain and demonstrates a complete loss of transactivation activity in yeast based functional studies (Chappuis PO et al. Int. J. Cancer, 1999 Dec;84:587-93; IARC TP53 database: Kato S et al. Proc Natl Acad Sci USA. 2003 Jul 8;100(14):8424-9). Although this specific alteration has not been previously described in the germline, another alteration at this same codon (p.C275Y) was reported in a classic Li Fraumeni family (Frebourg T et al. Am. J. Hum. Genet., 1995 Mar;56:608-15). Based on internal structural analysis, this variant significantly destabilizes the protein structure and is significantly more destabilizing than other known pathogenic variants in the region (Cho Y et al., Science 1994 Jul; 265(5170):346-55). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic. - |
