Loss-of-function variants in TP53 are known to be pathogenic (PMID: 20522432). For these reasons, this variant has been classified as Pathogenic. A frame-shift variant at this codon (Q52fs) has been reported in an individual affected with pediatric adrenocortical carcinoma (PMID: 25584008). It is unclear whether this is the same variant or not due to the limited information provided in the literature. This sequence change creates a premature translational stop signal (p.Gln52Leufs*4) in the TP53 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). -
