The c.7926dupT pathogenic mutation, located in coding exon 54 of the NF1 gene, results from a duplication of T at nucleotide position 7926, causing a translational frameshift with a predicted alternate stop codon (p.K2643*). This alteration was identified in one individual from a cohort of 521 German and Turkish patients with a clinical diagnosis of neurofibromatosis type I (Fahsold R et al. Am J Hum Genet, 2000 Mar;66:790-818). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
