The p.R1260C variant (also known as c.3778C>T), located in coding exon 48 of the COL1A1 gene, results from a C to T substitution at nucleotide position 3778. The arginine at codon 1260 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
Osteogenesis imperfecta type I Uncertain:1
This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 1260 of the COL1A1 protein (p.Arg1260Cys). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with COL1A1-related conditions. ClinVar contains an entry for this variant (Variation ID: 547232).
