Studies have shown that this variant is associated with skipping of exon 3 but is expected to preserve the integrity of the reading frame (PMID: 18546366). This variant has been observed in individual(s) with neurofibromatosis type 1 (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 527581). This variant is not present in population databases (ExAC no frequency). This sequence change affects a donor splice site in intron 3 of the NF1 gene. RNA analysis indicates that this variant induces altered splicing and likely results in a shortened protein product. -
