This sequence change replaces glutamine, which is neutral and polar, with lysine, which is basic and polar, at codon 162 of the NF1 protein (p.Gln162Lys). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NF1-related conditions. ClinVar contains an entry for this variant (Variation ID: 584926). The p.Q162K variant (also known as c.484C>A), located in coding exon 5 of the NF1 gene, results from a C to A substitution at nucleotide position 484. The glutamine at codon 162 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
