The p.Y95C variant (also known as c.284A>G), located in coding exon 2 of the FLCN gene, results from an A to G substitution at nucleotide position 284. The tyrosine at codon 95 is replaced by cysteine, an amino acid with highly dissimilar properties. In one study, this alteration was identified in a cohort of 21 hepatoblastoma patients (Aguiar T et al. Front Genet, 2022 Apr;13:858396). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 95 of the FLCN protein (p.Tyr95Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with hepatoblastoma (PMID: 35495172). ClinVar contains an entry for this variant (Variation ID: 460611). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt FLCN protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
