The p.Y408* pathogenic mutation (also known as c.1224T>A), located in coding exon 11 of the NF1 gene, results from a T to A substitution at nucleotide position 1224. This changes the amino acid from a tyrosine to a stop codon within coding exon 11. This alteration has been previously identified in an individual meeting diagnostic criteria of neurofibromatosis 1 (NF1) (Fahsold R et al. Am. J. Hum. Genet., 2000 Mar;66:790-818). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
