The c.1262_1263delGCinsAT variant (also known as p.S421N), located in coding exon 9 of the STK11 gene, results from a deletion of GC and insertion of AT between nucleotide positions 1262 and 1263. This results in the substitution of the serine residue for an asparagine residue at codon 421, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces serine with asparagine at codon 421 of the STK11 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373).
