This sequence change creates a premature translational stop signal (p.Trp1205Glyfs*32) in the NPHS1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 37 amino acid(s) of the NPHS1 protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with NPHS1-related conditions (PMID: 21415313). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 553267). This variant disrupts a region of the NPHS1 protein in which other variant(s) (p.Glu1207Lysfs*30) have been observed in individuals with NPHS1-related conditions (PMID: 33980730). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. -
