Variant summary: ETHE1 c.482G>A (p.Cys161Tyr) results in a non-conservative acid change located in the Metallo-beta-lactamase domain (IPR001279) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251474 control chromosomes (gnomAD). c.482G>A has been reported in the literature in an individual affected with Ethylmalonic Encephalopathy (Tiranti_2006). These data do not allow any conclusion about variant significance. At least one publication reports experimental evidence evaluating an impact on protein function, finding that the variant significantly impairs iron content, thermal stability, and catalytic activity (Kabil_2018). The following publications have been ascertained in the context of this evaluation (PMID: 16183799, 29980601). ClinVar contains an entry for this variant (Variation ID: 435095). Based on the evidence outlined above, the variant was classified as likely pathogenic. -
