The encoded protein is a component of the postsynaptic density at excitatory synapses, and may play a critical role in cytoskeletal and synaptic organization through the activation of selected ARF substrates including ARF1 and ARF6. Mutations in this gene have been implicated in nonsyndromic X-linked cognitive disability. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. The c.1676delC variant causes a frameshift starting with codon Proline 559, changes this amino acid to a Histidine residue, and creates a premature Stop codon at position 48 of the new reading frame, denoted p.Pro559HisfsX48. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.1676delC variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.1676delC as a pathogenic variant. - |
