The c.686T>G; p.Leu229Ter variant, to our knowledge, is not reported in the medical literature but is reported in ClinVar (Variation ID: 455925). Additionally, another variant (c.686T>A) that leads to the same p.Leu229Ter alteration has been reported in an individual with DMD and in a DMD carrier (Flanigan 2009, Takeshima 2010). The c.686T>G variant is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. This variant induces an early termination codon and is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay.
