Likely pathogenic, criteria provided, single submitter | clinical testing | Ambry Genetics | Dec 12, 2023 | The p.E501* variant (also known as c.1501G>T), located in coding exon 13 of the CHEK2 gene, results from a G to T substitution at nucleotide position 1501. This changes the amino acid from a glutamic acid to a stop codon within coding exon 13. This alteration occurs at the 3' terminus of the CHEK2 gene, is not expected to trigger nonsense-mediated mRNA decay, and only impacts the last 7.9% of the protein. However, premature stop codons are typically deleterious in nature and a significant portion of the protein is affected (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic. - |
