Variant summary: AGXT c.244G>A (p.Gly82Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251220 control chromosomes (gnomAD). c.244G>A has been observed in several homozygous individuals affected with Primary Hyperoxaluria Type 1 (van Woerden_2004). These data indicate that the variant is very likely to be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.245G>A, p.Gly82Glu), supporting the critical relevance of codon 82 to AGXT protein function. The following publication has been ascertained in the context of this evaluation (PMID: 15253729). ClinVar contains an entry for this variant (Variation ID: 2681157). Based on the evidence outlined above, the variant was classified as pathogenic. -
