Genes affected
AGXT (HGNC:341): (alanine--glyoxylate aminotransferase) This gene is expressed only in the liver and the encoded protein is localized mostly in the peroxisomes, where it is involved in glyoxylate detoxification. Mutations in this gene, some of which alter subcellular targetting, have been associated with type I primary hyperoxaluria. [provided by RefSeq, Jul 2008]

Primary hyperoxaluria Pathogenic:1
Likely pathogenic, criteria provided, single submitter | clinical testing | Women's Health and Genetics/Laboratory Corporation of America, LabCorp | Jul 19, 2024 | Variant summary: AGXT c.248A>G (p.His83Arg) results in a non-conservative amino acid change located in the Aminotransferase class V domain of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251192 control chromosomes. c.248A>G has been reported in the literature in at-least one individual affected with Primary Hyperoxaluria Type 1 (example: Williams_2009). Multiple publications have reported experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in <10% of normal activity (examples: Williams_2009, Pittman_2012, Messa-Torres_2013). The following publications have been ascertained in the context of this evaluation (PMID: 19479957, 24205397, 22923379). ClinVar contains an entry for this variant (Variation ID: 204085). Based on the evidence outlined above, the variant was classified as likely pathogenic. - |
