A heterozygous mutation in this gene was found in individuals with autosomal dominant distal hereditary motor neuronopathy type IID. There is a pseudogene for this gene on chromosome 4. This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 817 of the FBXO38 protein (p.Arg817His). This variant is present in population databases (rs183483408, gnomAD 0.05%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with FBXO38-related conditions. ClinVar contains an entry for this variant (Variation ID: 574698).
