This variant has been identified in 2/8654 East Asian chromosomes by the Exo me Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs1857021 48); however, its frequency is not high enough to rule out a pathogenic role. Co mputational prediction tools and conservation analyses of the amino acid change do not provide strong support for or against an impact to the protein. In summar y, the clinical significance of the p.Asp397Asn variant is uncertain. -
Variant summary: TMC1 c.1189G>A (p.Asp397Asn) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251422 control chromosomes. c.1189G>A has been reported in the literature in at least two individuals affected with Nonsyndromic Hearing Loss And Deafness, Type 7 (e.g., Yuan_2020, Xiang_2022). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 35761346, 31541171). ClinVar contains an entry for this variant (Variation ID: 229315).
