The leucine at codon 427 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the supporting evidence, this variant is unlikely to be causative of autosomal dominant SCN9A-related neuropathic pain syndromes; however, its contribution to the development of autosomal recessive congenital insensitivity to pain is uncertain. - |
