When cloned, it was discovered to be a human homolog of the E. coli mismatch repair gene mutS, consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with isoleucine at codon 135 of the MSH2 protein (p.Leu135Ile). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and isoleucine. -
