This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 537 of the KCNH2 protein (p.Arg537Trp). -

This variant has been reported in compound heterozygous state with a pathogenic truncation variant in an individual affected with a severe, childhood-onset long QT syndrome (PMID: 19184172). The proband's parents were asymptomatic heterozygous carriers. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
