This variant is found within a highly conserved region in the C-terminal cytoplasmic domain (a.a. 843-1159). Rare non-truncating variants in this region have been shown to be significantly overrepresented in individuals with long QT syndrome (PMID: 32893267). This variant has been shown to cause reduced channel current and accelerated rates of deactivation in a functional study with HEK293 cells (PMID: 31557540), while has also been shown to have no impact on channel current in another in vitro study (PMID: 34930020). This variant has been reported in two individuals affected with long QT syndrome (PMID: 17905336, 20850565), in an individual affected with complete atrioventricular block and torsades de pointes (PMID: 20386770), in an individual suspected of having epilepsy (PMID: 31696929), and in three individuals without arrhythmia (PMID: 34930020). This variant has been identified in 5/249490 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Although there is a suspicion for a pathogenic role, the available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |
