The p.R121Q variant (also known as c.362G>A), located in coding exon 2 of the SCN5A gene, results from a G to A substitution at nucleotide position 362. The arginine at codon 121 is replaced by glutamine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent with Brugada syndrome (Kapplinger JD et al. Heart Rhythm, 2010 Jan;7:33-46; Liu C et al. Forensic Sci Int, 2014 Mar;236:38-45; Berthome P et al. Heart Rhythm, 2019 Feb;16:260-267; Silva MA et al. Arq Bras Cardiol, 2021 Jul;117:19-22; Rucinski C et al. Mol Genet Genomic Med, 2022 Nov;10:e2046; Lu F et al. Orphanet J Rare Dis, 2022 Jan;17:2; Ambry internal data; external communication).
