This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 1786 of the SCN5A protein (p.Leu1786Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with long QT syndrome (PMID: 28412158; Invitae). ClinVar contains an entry for this variant (Variation ID: 692258). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Leu1786 amino acid residue in SCN5A. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 24599044).
