The serine at codon 1503 is replaced by proline, an amino acid with similar properties, and is located in the DIII/DIV interdomain linker region. This variant has been determined to be the result of a de novo mutation or germline mosaicism in one individual with prolonged QT syndrome (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. Based on internal structural analysis, this variant is more disruptive than known pathogenic variants (Shen H et al. Science, 2017 03;355:[Epub ahead of print]). In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic. - |
