This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 534 of the KCNH2 protein (p.Arg534Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of long QT syndrome (PMID: 16414944, 19716085; internal data). ClinVar contains an entry for this variant (Variation ID: 67221). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects KCNH2 function (PMID: 23546015). This variant disrupts the p.Arg534 amino acid residue in KCNH2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 9600240, 11668638, 18441445, 21308345, 22949429). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
