Variant summary: Variant summary: SCN5A c.89A>G (p.Glu30Gly) results in a non-conservative amino acid change in the encoded protein sequence. Three of three in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 248520 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.89A>G has been reported in the literature in individuals affected with clinical features of SCN5A-related conditions (examples: Kapplinger_2009 and Giudicessi_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Long QT Syndrome. At least one publication reports experimental evidence evaluating an impact on protein function. These results showed no damaging effect of this variant (Giudicessi_2020). The following publications have been ascertained in the context of this evaluation (PMID: 19716085, 32091595). ClinVar contains an entry for this variant (Variation ID: 68057). Experimental studies have shown that this missense change does not substantially affect SCN5A function (PMID: 32091595).
