This missense change has been observed in individuals with Brugada syndrome, clinical features of Brugada syndrome, and/or dilated cardiomyopathy (PMID: 12106943, 21273195, 30847665, 30847666). This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 1743 of the SCN5A protein (p.Gly1743Glu). This variant is not present in population databases (gnomAD no frequency). ClinVar contains an entry for this variant (Variation ID: 67968). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects SCN5A function (PMID: 16945804, 33131149). This variant disrupts the p.Gly1743 amino acid residue in SCN5A. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 12639704, 15023552, 22984773, 23420830, 25904541). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. -
