This alteration has been observed in multiple individuals with a personal and/or family history that is consistent with NF1-related disease (van Minkelen R et al. Clin Genet, 2014 Apr;85:318-27; D'Angelo F et al. Nat Med, 2019 Jan;25:176-187; Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
