Four of five in-silico tools predict a damaging effect of the variant on protein function. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 250874 control chromosomes (gnomAD). c.844G>A has been reported in the literature in multiple individuals affected with Phenylalanine Hydroxylase Deficiency (Phenylketonuria, e.g. Jeannesson-Thivisol_2015, Sterl_2013, Bayat_2016), including one homozygote (Bayat_2016). These data indicate that the variant is very likely to be associated with disease. One publication reports that the purified variant protein (from bacterial and cell free in vitro transcription-translation systems) had 1.3% and 0-4% enzymatic activity, respectively (Gjetting_2001).
