Caveolin proteins are proposed to be scaffolding proteins for organizing and concentrating certain caveolin-interacting molecules. Mutations identified in this gene lead to interference with protein oligomerization or intra-cellular routing, disrupting caveolae formation and resulting in Limb-Girdle muscular dystrophy type-1C (LGMD-1C), hyperCKemia or rippling muscle disease (RMD). Alternative splicing has been identified for this locus, with inclusion or exclusion of a differentially spliced intron. This missense change has been observed in individuals with autosomal dominant limb-girdle muscular dystrophy (PMID: 18671188, 19726876, 26185955; Invitae). ClinVar contains an entry for this variant (Variation ID: 31722). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Arg27 amino acid residue in CAV3. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 14633633, 15580566, 18930476, 19380584). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
