Genes affected
STRC (HGNC:16035): (stereocilin) This gene encodes a protein that is associated with the hair bundle of the sensory hair cells in the inner ear. The hair bundle is composed of stiff microvilli called stereocilia and is involved with mechanoreception of sound waves. This gene is part of a tandem duplication on chromosome 15; the second copy is a pseudogene. Mutations in this gene cause autosomal recessive non-syndromic deafness. [provided by RefSeq, Jul 2008]
CKMT1B (HGNC:1995): (creatine kinase, mitochondrial 1B) Mitochondrial creatine (MtCK) kinase is responsible for the transfer of high energy phosphate from mitochondria to the cytosolic carrier, creatine. It belongs to the creatine kinase isoenzyme family. It exists as two isoenzymes, sarcomeric MtCK and ubiquitous MtCK, encoded by separate genes. Mitochondrial creatine kinase occurs in two different oligomeric forms: dimers and octamers, in contrast to the exclusively dimeric cytosolic creatine kinase isoenzymes. Many malignant cancers with poor prognosis have shown overexpression of ubiquitous mitochondrial creatine kinase; this may be related to high energy turnover and failure to eliminate cancer cells via apoptosis. Ubiquitous mitochondrial creatine kinase has 80% homology with the coding exons of sarcomeric mitochondrial creatine kinase. Two genes located near each other on chromosome 15 have been identified which encode identical mitochondrial creatine kinase proteins. [provided by RefSeq, Jul 2008]

Autosomal recessive nonsyndromic hearing loss 16 Pathogenic:2
Likely pathogenic, no assertion criteria providedresearchDivision of Human Genetics, Children's Hospital of PhiladelphiaMay 23, 2015The heterozygous variant was identified in the STRC gene (c.4701+1G>A) is considered likely pathogenic. This variant was previously been seen in 10 individuals in ExAC and has not been previously published. Available splice predictors () predict a complete loss of the splice site with a potential shifting of the splice by one nucleotide which would result in a frame shift. -

Rare genetic deafness Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineNov 03, 2022The c.4701+1G>A variant in STRC has been previously reported in 1 individual with hearing loss who carried a second STRC variant in trans (Sheppard et al., PMID: 29907799) and another individual with hearing loss that carried a pathogenic STRC deletion. This variant has also been identified in 8/16512 South Asian and 2/66724 European chromosomes by the Exome Aggregation Consortium (http://exac.broadinstitute.org/; dbSNP rs199839039). Although this variant has been seen in the general population, its frequency is low enough to be consistent with a recessive carrier frequency. This variant occurs in the invariant region (+1/2) of the splice consensus sequence and is predicted to cause altered splicing leading to an abnormal or absent protein. In summary, this variant meets our criteria to be classified as pathogenic for autosomal recessive hearing loss based on the predicted impact. ACMG/AMP criteria applied: PSV1_Strong, PM3_Strong. -
