This reaction uses calcium as a cofactor and plays an important role in the intracellular transduction of many extracellular signals. The p.N1178S variant (also known as c.3533A>G), located in coding exon 32 of the PLCB1 gene, results from an A to G substitution at nucleotide position 3533. The asparagine at codon 1178 is replaced by serine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be benign and tolerated by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
