The glycine at codon 723 is replaced by valine, an amino acid with dissimilar properties. This alteration was found in heterozygous state in one individual who reportedly had cystic fibrosis; however, clinical information was limited and no second CFTR alteration was detected (Zitkiewicz E et al. PLoS ONE, 2014 Feb;9:e89094). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 251230 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2168G>T has been reported in the literature in the heterozygous state in at least one individual affected with Cystic Fibrosis without a second reported variant (e.g. Zietkiewicz_2014). This report does not provide unequivocal conclusions about association of the variant with Chronic Pancreatitis Risk or other CFTR-related disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 25735457, 24586523). ClinVar contains an entry for this variant (Variation ID: 53447).
