TECTA (HGNC:11720): (tectorin alpha) The tectorial membrane is an extracellular matrix of the inner ear that contacts the stereocilia bundles of specialized sensory hair cells. Sound induces movement of these hair cells relative to the tectorial membrane, deflects the stereocilia, and leads to fluctuations in hair-cell membrane potential, transducing sound into electrical signals. Alpha-tectorin is one of the major noncollagenous components of the tectorial membrane. Mutations in the TECTA gene have been shown to be responsible for autosomal dominant nonsyndromic hearing impairment and a recessive form of sensorineural pre-lingual non-syndromic deafness. The p.Val1494Ala variant (rs200544452) has not been reported in the medical literature or gene specific variation databases but has been reported to ClinVar as a variant of uncertain significance (Variation ID: 180103). This variant is listed in the genome Aggregation Database (gnomAD) with an overall population frequency of 0.01 percent (identified on 36 out of 275,330 chromosomes). The valine at position 1494 is highly conserved, up to Tetraodon (considering 11 species) (Alamut v.2.10.0) and computational analyses of the effects of the p.Val1494Ala variant on protein structure and function provide conflicting results (SIFT: tolerated, MutationTaster: disease causing, PolyPhen-2: probably damaging). Altogether, there is not enough evidence to classify the p.Val1494Ala variant with certainty. -

The p.Val1494Ala variant in TECTA has not been previously reported in individual s with hearing loss, but has been identified in 6/66584 European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs 200544452). Although this variant has been seen in the general population, its f requency is not high enough to rule out a pathogenic role. Computational predict ion tools and conservation analyses suggest that this variant may impact the pro tein, though this information is not predictive enough to determine pathogenicit y. In summary, the clinical significance of p.Val1494Ala variant is uncertain. -
