The p.Arg2360Cys variant in PLEC has not been previously reported in individuals with myopathy, but has been identified in 6/15984 South Asian chromosomes by th e Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs20 0620255). Computational tools predict that this variant may impact the protein; however, arginine (Arg) at position 2360 is not well conserved in evolution with some species (black flying fox, megabat) having the variant amino acid, raising the possibility that this change may be tolerated. In summary, the clinical si gnificance of the p.Arg2360Cys variant is uncertain. -
