The threonine at codon 1373 is replaced by alanine, an amino acid with similar properties. This alteration was initially reported in a Finnish male athlete with ventricular tachycardia and an enlarged right ventricle (Lahtinen AM, Heart Rhythm 2011 Aug; 8(8):1214-21; Lahtinen AM, Ann. Med. 2013 Jun; 45(4):328-35). This alteration has been detected in a cohort referred for arrhythmogenic right ventricular cardiomyopathy (ARVC) genetic testing and in a Finnish cohort of sudden death victims with coronary artery disease; however, clinical details were limited (Walsh R et al. Genet. Med., 2017 02;19:192-203; V&auml;h&auml;talo JH et al. Front Cardiovasc Med. 2021 Jan;8:755062). This variant has also been detected in Finnish control alleles, and in several individuals from an exome cohort without know ARVC; however, details were limited (Haggerty CM et al. Genet Med. 2017 11;19(11):1245-1252; V&auml;h&auml;talo JH et al. Front Cardiovasc Med. 2021 Jan;8:755062). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
