CBS (HGNC:1550): (cystathionine beta-synthase) The protein encoded by this gene acts as a homotetramer to catalyze the conversion of homocysteine to cystathionine, the first step in the transsulfuration pathway. The encoded protein is allosterically activated by adenosyl-methionine and uses pyridoxal phosphate as a cofactor. Defects in this gene can cause cystathionine beta-synthase deficiency (CBSD), which can lead to homocystinuria. This gene is a major contributor to cellular hydrogen sulfide production. Multiple alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Feb 2016]

This frequency is not significantly higher than estimated for a pathogenic variant in CBS causing Homocystinuria (8.8e-05 vs 0.003), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1411G>A in individuals affected with Homocystinuria and no experimental evidence demonstrating its impact on protein function have been reported. The G471R variant has not been published as pathogenic in association with homocystinuria or been reported as benign to our knowledge. The G471R variant is observed in 5/11554 (0.043%) alleles from individuals of Latino background in the Exome Aggregation Consortium (ExAC) dataset, though no individuals were reported to be homozygous (Lek et al., 2016). The G471R variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species and in silico analysis predicts this variant is probably damaging to the protein structure/function. -
