The PCNT c.427C>A variant is predicted to result in the amino acid substitution p.Arg143Ser. To our knowledge, this variant has not been reported in the literature. An alternate substitution impacting the same amino acid (p.Arg143His) has been reported to have occurred de novo in an individual with autism spectrum disorder (Table S2, Iossifov et al. 2014. PubMed ID: 25363768), although it should also be noted that the p.Arg143His substitution was reported on 0.37% of alleles in individuals of Ashkenazi Jewish descent in gnomAD (http://gnomad.broadinstitute.org/variant/21-47754471-G-A), which may be too common to be a primary cause of disease. The c.427C>A (p.Arg143Ser) variant is reported in 0.012% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/21-47754470-C-A). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
