The asparagine at codon 796 is replaced by histidine, an amino acid with similar properties. This alteration has been reported in the germline of 2 of 8,920 ethnically matched normal population control subjects and 1 of 516 samples from a study of chronic lymphocytic leukemia patients of European descent (Tiao G et al. Leukemia, 2017 10;31:2244-2247). In another study, this alteration was identified in a female breast cancer patient diagnosed at age 35 (Feliubadaló L et al. Clin Chem, 2021 03;67:518-533). This alteration was also identified in the germline of a patient with cutaneous melanoma diagnosed at age 67; functional studies showed reduced protein expression and tumor LOH (Pastorino L et al. Int J Mol Sci, 2022 Dec;23). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
