This variant is present in population databases (no rsID available, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with SPRED1-related conditions. The p.V229I variant (also known as c.685G>A) is located in coding exon 7 of the SPRED1 gene. The valine at codon 229 is replaced by isoleucine, an amino acid with highly similar properties. This change occurs in the first base pair of coding exon 7. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
