Genes affected
DSC2 (HGNC:3036): (desmocollin 2) This gene encodes a member of the desmocollin protein subfamily. Desmocollins, along with desmogleins, are cadherin-like transmembrane glycoproteins that are major components of the desmosome. Desmosomes are cell-cell junctions that help resist shearing forces and are found in high concentrations in cells subject to mechanical stress. This gene is found in a cluster with other desmocollin family members on chromosome 18. Mutations in this gene are associated with arrhythmogenic right ventricular dysplasia-11, and reduced protein expression has been described in several types of cancer. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Feb 2015]

Likely benign, criteria provided, single submitterclinical testingGeneDxApr 20, 2020Reported in an individual with a personal and family history of DCM; this individual also harbored a frameshift variant in the TTN gene that was reported as likely pathogenic by the authors (Pugh et al., 2014); Reported as a non-segregating variant in a case of sporadic, pediatric DCM (Long et al., 2017); Reported in ClinVar as a variant of uncertain significance and as a likely benign variant; one clinical laboratory reports observing this variants in two adults with DCM, both of whom harbored another likely pathogenic variant (ClinVar Variant ID#46170; SCV000063095.5; Landrum et al., 2016); In silico analysis supports that this missense variant does not alter protein structure/function; This variant is associated with the following publications: (PMID: 23861362, 24503780, 29367541, 31737537) -

Uncertain significance, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineJul 03, 2014The Ile577Val variant in DSC2 has been identified by our laboratory in 2 Caucasi an adults with DCM (including this family), both of whom carried another likely pathogenic variant. This variant has also been identified in 1/8600 European Ame rican chromosomes by the NHLBI Exome Sequencing Project (http://evs.gs.washingto n.edu/EVS/; dbSNP rs201845641) and was reported in 1/186 controls in an unpublis hed study (ARVD/C Database, http://www.arvcdatabase.info). Computational predict ion tools and conservation analysis do not provide strong support for or against an impact to the protein. In summary, additional information is needed to fully assess the clinical significance of the Ile577Val variant. -
